Diffusion Coefficient Measurement

Diffusion coefficient measurement determines how quickly molecules or particles spread through a medium, providing a quantitative description of transport in biological systems. It typically tracks concentration changes, displacement, or signal fluctuations over time and relates them to diffusion using Fick’s laws or, for random motion, mean-squared displacement; the coefficient depends on temperature, viscosity, particle size, and interactions with the environment. In biology, these measurements characterize membrane permeability, intracellular mobility, protein dynamics, and drug transport. The resulting data help distinguish free from hindered diffusion and connect molecular movement with signaling, metabolism, and cellular organization.

In this article we will describe the procedure for measuring diffusion coefficients using multi-photon fluorescence recovery after photobleaching. We will begin by aligning the laser along the optical path to the sample and determining the proper experimental parameters, then continue generating and finally fitting fluorescence recovery curves.

By ideal inviscid potential flow theory, the pressure distribution around a cylinder is vertically symmetric. The pressure distribution upstream and downstream of the cylinder is also symmetric, which results in a zero-net drag force.
