Drosophila Leg Dissection

Drosophila leg dissection is a laboratory technique for isolating and exposing the fly’s segmented leg to examine its anatomy, development, and function. Researchers anesthetize the fly, secure the specimen, and use fine instruments under a stereomicroscope to remove surrounding cuticle and tissue while preserving structures such as joints, muscles, sensory organs, and nerves. The preparation supports biological studies of appendage patterning, neuromuscular organization, sensory biology, and movement. By enabling direct visualization and labeling of specific tissues, Drosophila leg dissection helps connect genetic or molecular changes with cellular structure and locomotor phenotypes.

The limiting factor in the use of the adult Drosophila eye to study neurodegeneration and cell biology is the difficult imaging of intracellular processes. We describe the dissection of single ommatidia to generate a bona-fide primary neuronal cell culture, which can be subject to drug treatment and advanced imaging.
