Engineered Rna Target

An engineered RNA target is a deliberately designed RNA sequence or structural element that enables researchers to direct, monitor, or regulate a specific biological process. Its activity typically depends on complementary base pairing with a guide RNA or nucleic acid probe, or on selective recognition by an RNA-binding protein, allowing the target to be detected, cleaved, stabilized, or translated under defined conditions. In biology, engineered RNA targets support gene regulation, RNA interference, molecular diagnostics, and synthetic biology, where controlled sequence design can improve specificity and measurement. These targets also help researchers investigate RNA structure, gene expression, and programmable cellular responses.

RNA Pull-down Procedure to Identify RNA Targets of a Long Non-coding RNA

This RNA pull-down method allows identifying the RNA targets of a long non-coding RNA (lncRNA). Based on the hybridization of home-made, designed anti-sense DNA oligonucleotide probes specific to this lncRNA in an appropriately fixed tissue or cell line, it efficiently allows the capture of all RNA targets of the lncRNA.

Ratiometric bimolecular beacons (RBMBs) can be used to image single engineered RNA transcripts in living cells. Here, we describe the preparation and purification of RBMBs, delivery of RBMBs into cells by microporation and fluorescent imaging of single RNA transcripts in real-time.

We describe a targeted RNA sequencing-based method that includes preparation of indexed cDNA libraries, hybridization and capture with custom probes and data analysis to interrogate selected transcripts for gene expression, mutations, and gene fusions. Targeted RNAseq permits cost-effective, rapid evaluation of selected transcripts on a desktop sequencer.

Helicase Activity Measurement of a Target Protein Using Biotin-Labeled RNA Duplexes

In this video, we demonstrate the procedure to determine the helicase activity of a target protein to unwind the biotin-labeled dsRNA substrate. The activity of the enzyme was identified by analyzing the electrophoretic mobility shift, followed by a chemiluminescence assay using chemiluminescent enzyme-conjugated streptavidin.
