Epithelial-like Cells

Epithelial-like cells are cultured cells that resemble epithelial cells in shape and organization, typically forming closely packed sheets, but their appearance alone does not confirm their tissue origin or full epithelial function. Their characteristic morphology arises through cell spreading, cell-cell adhesion, substrate attachment, and the development of organized contacts that may produce apical-basal polarity. In biology, these cells provide useful models for studying tissue organization, barrier formation, wound closure, and cellular responses to experimental conditions or compounds. Careful analysis of morphology, junctions, and molecular markers helps distinguish epithelial-like behavior from a definitive epithelial identity.

We present a protocol that combines cell isolation and whole-cell patch-clamp recording to measure the electrical properties of the primary dissociated epithelial cells from the rat cauda epididymides. This protocol allows for investigation of the functional properties of primary epididymal epithelial cells to further elucidate the physiological role of the epididymis.

We present here a cell culture method for inducing mesenchymal-epithelial transitions (MET) in sarcoma cells based on combined ectopic expression of microRNA-200 family members and grainyhead-like 2 (GRHL2). This method is suitable for better understanding the biological impact of phenotypic plasticity on cancer aggressiveness and treatments.
