Fas System

The Fas system is a cell-signaling pathway that regulates programmed cell death, helping maintain tissue balance and eliminate unwanted or damaged cells. When Fas ligand binds the Fas receptor on a target cell, receptor clustering recruits adaptor proteins to form the death-inducing signaling complex, which activates initiator caspase-8 and downstream executioner caspases. This extrinsic apoptosis pathway can also amplify cell death through mitochondrial signaling, linking external death signals to intracellular responses. In biology, the Fas system is essential for immune-cell homeostasis, tolerance, and removal of infected or abnormal cells, while dysregulation can contribute to autoimmune disease, immunodeficiency, and cancer.

An Ex Vivo Technique to Induce Neuronal Death via Oligodendrocyte-Specific CD8+ T Cells

2025

This video demonstrates an ex vivo technique to induce brain cell death mediated through cytotoxic CD8+ T cells. Upon obtaining brain slices, activated oligodendrocyte-directed CD8+ T cells are transferred onto the slices to target the myelin sheath and oligodendrocytes. Post-incubation, the damage to myelin and neuronal death can be assessed histologically in the regions of interest.

An Assay to Study the Impact of Extracellular Matrix Stiffness on Bacterial Infection

2025

This video demonstrates an assay to study the effect of extracellular matrix (ECM) stiffness on bacterial infection of adherent cells. Polyacrylamide hydrogels of varying stiffness, overlaid with collagen, are layered in a multi-well plate to mimic the stiffness of physiological ECM. Upon overlaying human endothelial cells on the hydrogels and infecting the cells with Listeria monocytogenes bacteria engineered to express a fluorescent protein once intracellular, flow cytometry is used to...
