Fatty Acid Oxidation

Fatty acid oxidation is the metabolic process that breaks down fatty acids to release energy, supporting cellular function during fasting, exercise, and increased energy demand. In the mitochondria, fatty acids are activated and transported into the matrix, where beta-oxidation repeatedly removes two-carbon units as acetyl-CoA while generating NADH and FADH2; these products fuel the citric acid cycle and electron transport chain to produce ATP. This pathway is central to energy metabolism in biology and helps explain how cells use stored lipids. Studying fatty acid oxidation also supports research into metabolic disorders, exercise physiology, nutrition, and mitochondrial disease.

De novo lipogenesis and β-fatty acid oxidation constitute key metabolic pathways in hepatocyte, pathways that are perturbed in several metabolic disorders, including fatty liver disease. Here we demonstrate isolation of mouse primary hepatocytes and describe quantification of β-fatty acid oxidation and lipogenesis.

This protocol details an in vitro assay for measuring cellular lipid uptake in endothelial cells upon stimulation with BODIPY-C12 and BODIPY-C16 fluorescent analogs of long-chain and very long-chain saturated fatty acids. This method is efficient and adaptable to other cell types, offering a useful approach for studying lipid metabolism.

We propose a protocol to identify fatty acids without the need to purify them. It combines information on the retention times with the mass spectra of three types of fatty acid derivatives: fatty acid methyl esters (FAMEs), 4,4-dimethyl oxazoline derivatives (DMOX), and 3-pyridylcarbinyl esters (picolinyl).
