Fluorescence Assisted Cell Sorter

A Fluorescence Assisted Cell Sorter is a flow cytometry instrument that identifies and physically separates cells according to fluorescence and other measurable properties, enabling researchers to isolate defined cell populations. As cells pass individually through laser beams, fluorescently labeled molecules emit light detected by the instrument; the system then places cells into droplets and uses electrostatic charging and deflection to direct selected droplets into separate collection tubes. This method supports analysis and purification of immune, stem, tumor, and genetically modified cells. Sorted populations can be used for molecular profiling, functional assays, culture, and investigations of cellular heterogeneity in biology.

The manuscript describes a method of phonon-assisted quasi-resonant fluorescence spectroscopy that incorporates both laser-limited resolution and photoluminescence (PL) spectroscopy. This method utilizes optical phonons to provide linewidth-limited resolution spectra of atom-like semiconductor structures in the energy domain. The method is also easily realized with a single spectrometer optical spectroscopy setup.

Liquid-grown Streptomyces cultures are characterized by mycelial pellets that are heterogeneous in size. We here describe a method to analyze and sort such pellets in a high-throughput manner. These pellets can be used for further analyses, which will provide leads to understand and control growth heterogeneity.

A method for isolating specific cell types from plant material is demonstrated. This technique employs transgenic marker lines expressing fluorescent proteins in particular cell types, cellular dissociation and Fluorescence Activated Cell Sorting. Additionally, a growth setup is established here that facilitates treatment of Arabidopsis thaliana seedlings prior to cell sorting.
