DNA and RNA analysis comprises laboratory methods used to characterize nucleic acids, revealing genetic information, gene expression, and molecular changes in cells and organisms. These methods typically isolate DNA or RNA, convert or amplify target sequences when needed, and measure them through techniques such as polymerase chain reaction, reverse transcription, electrophoresis, hybridization, or sequencing. In biology, the resulting data help identify mutations, compare genomes, quantify transcripts, track pathogens, and investigate developmental or disease-related processes. Careful sample handling and appropriate controls are essential because degradation, contamination, and amplification bias can alter results and affect interpretation.

Assessment of DNA Contamination in RNA Samples Based on Ribosomal DNA

Here, we present a protocol for tracing genomic DNA (gDNA) contamination in RNA samples. The presented method utilizes primers specific for the internal transcribed spacer region (ITS) of ribosomal DNA (rDNA) genes. The method is suited for reliable and sensitive detection of DNA contamination in most eukaryotes and prokaryotes.

Here we present a new approach to identify plant viruses with double-strand DNA genomes. We use standard methods to extract DNA and RNA from infected leaves and carry out next-generation sequencing. Bioinformatic tools assemble sequences into contigs, identify contigs representing virus genomes and assign genomes to taxonomic groups.
