Fluorescence Signal Measurement

Fluorescence signal measurement is a technique for detecting and quantifying light emitted by fluorescent molecules after they absorb light at a shorter wavelength, providing information about biological structures, molecules, and processes. In a typical measurement, excitation light promotes a fluorophore to a higher-energy state, and the fluorophore releases part of that energy as longer-wavelength emission; instruments measure the resulting signal as intensity, often relative to a control or calibration standard. In biology, this approach supports microscopy, protein and nucleic acid assays, cell labeling, and reporter-based studies of gene expression or signaling. Careful control of background fluorescence, photobleaching, and measurement conditions improves accuracy and reproducibility.

Phagosomal pH influences phagosome maturation, oxidant production, phagosomal killing as well as antigen presentation. Here we describe a ratiometric method for measuring time-course and endpoint pH changes in individual phagosomes in living phagocytes using fluorescence microscopy.
