Fluorescent Protein Localization

Fluorescent protein localization is a biological imaging method used to determine where specific proteins reside within living or fixed cells, revealing how spatial organization supports cellular function. Researchers genetically fuse a protein of interest to a fluorescent reporter, such as green fluorescent protein, then use excitation light and fluorescence microscopy to detect the emitted signal; targeting sequences and cellular conditions can help preserve normal distribution. This approach maps proteins to organelles, membranes, or other cellular regions and can track their movement over time. In biology, fluorescent protein localization supports studies of cell signaling, trafficking, development, and disease by linking protein position with activity.

The precise localization of Golgi residents is essential for understanding the cellular functions of the Golgi. However, conventional optical microscopy is unable to resolve the sub-Golgi structure. Here we describe the protocol for a conventional microscopy based super-resolution method to quantitatively determine the sub-Golgi localization of a protein.
