Fluorescent Secondary Antibodies

Fluorescent secondary antibodies are labeled immunoglobulins that detect primary antibodies, enabling researchers to visualize or measure specific proteins and other targets in biological samples. They work through selective binding between the secondary antibody and the host species or antibody class of the primary antibody; attached fluorophores then emit light at characteristic wavelengths after excitation, converting antibody binding into a detectable signal. This indirect approach can amplify signal because multiple secondary antibodies may bind one primary antibody and provides flexibility across experiments. In immunofluorescence microscopy, flow cytometry, and fluorescence-based assays, these reagents support protein localization, cell phenotyping, and comparative analysis of biological samples.

We described a procedure for the disaggregation of colorectal cancer (CRC) to produce viable single cells, which are then captured on customized antibody microarrays recognizing surface antigens (DotScan CRC microarray). Sub-populations of cells bound to the microarray can be profiled by fluorescence multiplexing using monoclonal antibodies tagged with fluorescent dyes.
