Gfp-tagged Marker

A GFP-tagged marker is a biological labeling tool in which green fluorescent protein (GFP) is genetically fused to a protein or linked to a regulatory sequence, enabling researchers to visualize specific cells, structures, or gene activity. When illuminated with light of the appropriate wavelength, GFP absorbs energy and emits green fluorescence, allowing the tagged target to be detected by fluorescence microscopy. In biology, GFP-tagged markers help track protein localization, monitor cellular processes, assess gene expression, and follow changes in living cells over time. These applications support research in cell biology, developmental biology, genetics, and disease mechanisms.

Source: Bouillier, C. et. al., Amplification, and Titration of Recombinant Respiratory Syncytial Viruses. J. Vis. Exp. (2019)This video demonstrates the generation of a green fluorescent protein (GFP)-expressing recombinant respiratory syncytial virus using a T7-based reverse genetics system. The co-transfection of plasmids encoding viral proteins and a GFP-tagged genome leads to protein expression and initiates virus rescue.

Source: Naji, A., et al. Studying Oxidative Stress Caused by the Mitis Group Streptococci in Caenorhabditis elegans. J. Vis. Exp. (2019)This video demonstrates the use of green fluorescent protein-tagged SKN-1 in C. elegans to monitor oxidative stress by tracking green fluorescence. Worms exposed to S. gordonii show strong nuclear fluorescence, confirming pathogen-induced stress, while those fed E. coli display diffuse green fluorescence in the cytoplasm, indicating minimal SKN-1 activation.
