Gpcr Classes

GPCR classes are groups of G protein-coupled receptors categorized by sequence, structure, ligand preference, and signaling features, making them essential for understanding how cells sense and respond to extracellular cues. These receptors typically span the cell membrane seven times; ligand binding changes their conformation, enabling interaction with heterotrimeric G proteins and regulation of intracellular effectors such as adenylyl cyclase or phospholipase C. Major groups, including classes A, B, C, and F, respond to diverse molecules ranging from hormones and neurotransmitters to sensory stimuli. Their classification supports studies of cell communication, physiology, disease mechanisms, and drug development, since many therapeutically important targets are GPCRs.

GPCR Desensitization

2023

G protein-coupled receptor (GPCR) signaling plays a crucial role in cell functioning. GPCR desensitization is an equally essential process. It allows cells to respond to changing environments and regain sensitivity to new stimuli while preventing unnecessary stimulation when no longer needed. Prolonged exposure to stimuli leads to GPCR desensitization. It involves blocking the receptors from binding and activating additional G proteins. This inhibits activation of downstream effectors, thereby...

Simple methods to detect the selective activation of G proteins by G protein-coupled receptors remain an outstanding challenge in cell signaling. Here, Fӧrster resonance energy transfer (FRET) biosensors have been developed by pairwise tethering a GPCR to G protein peptides to probe conformational changes at controlled concentrations in live cells.

Antibodies, also known as immunoglobulins, are produced by B cells in response to foreign substances, such as bacteria and viruses. These proteins are critical for recognizing and neutralizing these substances, protecting the body from potential harm. The basic structure of an antibody consists of four protein chains: two identical heavy chains and two identical light chains. These chains are held together by disulfide bonds and other non-covalent interactions, forming a Y-shaped structure.

Drugs can be classified according to their chemical composition or their intended therapeutic application. For instance, anti-infective agents that possess the ability to eliminate pathogens or suppress their growth and reproduction can be grouped based on the organisms they target or their chemical structure. Furthermore, drugs can be divided into prescription, nonprescription, or controlled substances. Prescription medications, such as antibiotics, require oversight from a licensed healthcare...
