Gut Barrier Integrity

Gut barrier integrity is the capacity of the gastrointestinal lining to regulate what passes between the gut lumen and the body while blocking harmful substances and pathogens. It depends on mucus, intestinal epithelial cells, antimicrobial factors, immune defenses, and tight junction proteins that control paracellular permeability between adjacent cells. Disruption of this coordinated barrier can permit unwanted molecules or microbes to cross the epithelium, promoting local inflammation and altered host-microbe interactions. Studying gut barrier integrity helps researchers investigate intestinal physiology, nutrition, infection, inflammatory disease, and potential therapies that preserve or restore epithelial function.

2026

This article describes: (1) oral gavage delivery of test molecules to neonatal mice as early as DOL 6; and (2) ex vivo evaluation of colonic epithelial barrier function in biopsies from DOL 10 to weaning (DOL 20) pups by assessing paracellular and transcellular permeability with Ussing chambers.

In the present study, fluorescein-isothiocyanate-labeled (FITC) dextran is administered to mice via oral gavage to evaluate intestinal permeability both in vivo and in plasma and fecal samples. As gut barrier function is affected in many disease processes, this direct and quantitative assay can be used in diverse research areas.

Assessing the Role of Gut Microbiota in Modulating Hepatitis B Infection in Mice

Take a control mouse with a normal gut microbiota and an experimental mouse with depleted gut microbiota. The depletion compromises the integrity of the gut barrier. This enables bacteria to translocate through the blood vessel into the liver tissue.Inject both mice with the hepatitis B virus or HBV plasmid to induce an HBV infection in the liver tissue. In the control mouse, the T cell detects an infected liver cell through the interaction of its receptors with viral antigens, and produces...

Assessing Blood-Brain Barrier Integrity in a Laser-Induced Brain Injury Rat Model

Source: Kuts, R. et al. Laser-Induced Brain Injury in the Motor Cortex of Rats. J. Vis. Exp. (2020)This video demonstrates assessing blood-brain barrier (BBB) integrity following laser-induced brain injury in rats. A tracking dye is injected into the tail vein, allowing it to circulate to the brain and leak into the tissue through the damaged BBB. The brain is then extracted and processed to release the dye. The fluorescence intensity of the dye is measured to quantify the extent of...

Assay for Blood-brain Barrier Integrity in Drosophila melanogaster

2019

Blood-brain barrier integrity is critical for nervous system function. In Drosophila melanogaster, the blood-brain barrier is formed by glial cells during late embryogenesis. This protocol describes methods to assay for blood-brain barrier formation and maintenance in D. melanogaster embryos and third instar larvae.
