Heart Tissue Digestion

Heart tissue digestion is a laboratory method that breaks down cardiac tissue to release individual cells for biological study. Enzymes such as collagenase and proteases degrade the extracellular matrix, while controlled mechanical agitation and carefully maintained temperature and solution conditions help preserve cell viability. The resulting cell suspension can contain cardiomyocytes, fibroblasts, endothelial cells, and other cardiac cell types, depending on the tissue source and protocol. Researchers use this technique to examine cardiac structure and function, establish primary cultures, investigate development and disease, and evaluate cellular responses to drugs or injury.

Source: Brown, A. O., et al. Visualization of Streptococcus pneumoniae within Cardiac Microlesions and Subsequent Cardiac Remodeling. J. Vis. Exp. (2015).This video demonstrates the microscopic visualization of bacterial microlesions in mouse heart tissue. The heart of a mouse infected with Streptococcus pneumoniae is harvested, washed, and fixed before being processed to obtain coronal sections. These sections are stained with hematoxylin and eosin and examined under a microscope to visualize...
