Hemolymph Collection

Hemolymph collection is a biological sampling technique used to obtain the circulating fluid of invertebrates, including insects, crustaceans, and mollusks, for physiological and biochemical analysis. Researchers access the hemocoel or a hemolymphatic vessel with a fine needle, capillary, or micropipette, often using controlled restraint and anticoagulants to limit clotting and preserve sample integrity. Collected hemolymph can be examined for hemocytes, proteins, metabolites, hormones, pathogens, and immune activity. In biology, this method supports studies of invertebrate immunity, development, nutrition, toxicology, and disease, while enabling comparisons between experimental treatments and physiological states.

The collection of infected tick hemolymph, salivary glands, and saliva is important to study how tick-borne pathogens cause disease. In this protocol we demonstrate how to collect hemolymph and salivary glands from feeding Ixodes scapularis nymphs. We also demonstrate saliva collection from female I. scapularis adults.
