Hemolymph Injection

Hemolymph injection is a laboratory technique used to introduce substances directly into the circulatory fluid of arthropods such as insects, enabling controlled study of internal biological responses. A fine needle passes through the cuticle into the hemocoel, depositing a measured volume that mixes with hemolymph and is carried to tissues by circulation, thereby bypassing the digestive tract. Researchers use this method to deliver pathogens, toxins, hormones, or nucleic acids, then measure survival, immune defense, infection progression, or gene function. In biology, hemolymph injection supports studies of host-pathogen interactions, innate immunity, insect physiology, and pest control, while careful control of dose, injection site, and handling improves reproducibility.

Conserved insulin signaling pathways found in the fruit fly Drosophila melanogaster make this organism a potential tool for modeling metabolic disorders including type II diabetes. To this end, it is critical to establish physiological assays to effectively measure systemic insulin action in peripheral glucose disposal in the adult fly.

Cuticle Disruption: A Method to Collect Hemolymph from Drosophila Larvae

The collection of infected tick hemolymph, salivary glands, and saliva is important to study how tick-borne pathogens cause disease. In this protocol we demonstrate how to collect hemolymph and salivary glands from feeding Ixodes scapularis nymphs. We also demonstrate saliva collection from female I. scapularis adults.
