Heparin Acrylic Beads

Heparin acrylic beads are porous polymer particles bearing immobilized heparin, a sulfated polysaccharide that binds many proteins involved in biology and medicine. Their negatively charged heparin groups interact electrostatically and through structural recognition with positively charged regions on proteins, allowing selective capture under controlled buffer and salt conditions. In biotechnology, these beads support affinity chromatography for separating and purifying growth factors, coagulation proteins, and other heparin-binding molecules from complex mixtures. They also provide a surface for studying protein interactions and can help concentrate or deliver bioactive factors, making them useful in biochemical research, diagnostics, and tissue-engineering investigations.

This video demonstrates two methods for preparing and placing beads, which have been coated with growth factor, on explants of the developing cerebral cortex. These beads can be used to induce spatially restricted gene expression on developing neural tissue such as forebrain explants. Methods are given for using both Affi-Gel beads and heparin acryllic beads.

Self-assembled polyelectrolyte complexes (PEC) fabricated from heparin and protamine were deposited on alginate beads to entrap and regulate the release of osteogenic growth factors. This delivery strategy enables a 20-fold reduction of BMP-2 dose in spinal fusion applications. This article illustrates the benefits and fabrication of PECs.
