Heterochromatin Proteins

Heterochromatin proteins are DNA- and chromatin-associated factors that organize compact, relatively inactive regions of the genome and help preserve genome stability. In many systems, proteins such as heterochromatin protein 1 recognize methylated histone H3 at lysine 9, bind modified nucleosomes, and recruit additional chromatin-modifying enzymes, promoting nucleosome compaction and transcriptional repression. These proteins regulate repetitive DNA, centromeres, telomeres, and developmental gene programs while limiting inappropriate recombination and transposable-element activity. Studying their interactions with histones, DNA, and other nuclear factors helps explain epigenetic inheritance and provides insight into how disrupted chromatin organization contributes to disease.
