Double Ligation Technique

The Double Ligation Technique is a molecular biology method that joins DNA fragments to construct recombinant molecules, often by combining an insert with a plasmid vector. In a typical procedure, restriction enzymes generate defined DNA ends, complementary overhangs align the fragments, and DNA ligase forms phosphodiester bonds to seal both junctions; using two distinct end sequences can promote directional insertion and reduce vector self-ligation. This approach supports plasmid construction, gene cloning, and the assembly of expression or reporter constructs, providing a reliable foundation for studying gene function and producing engineered DNA for downstream biological experiments.
