High Density Cultures

High density cultures are laboratory systems designed to grow a large number of cells or microorganisms within a limited volume, increasing biomass or product yield. As cell concentration rises, nutrient and oxygen consumption intensify while metabolic waste accumulates, so controlled feeding, aeration, mixing, or perfusion may be used to maintain suitable growth conditions. These cultures support studies of microbial physiology, cell metabolism, and population behavior, and they are important for producing vaccines, enzymes, therapeutic proteins, and other biological products. Understanding how cells respond to crowding and resource limitations helps researchers improve culture efficiency, reproducibility, and scale-up from laboratory systems to bioprocessing applications.

Generating an Ultra-Low-Density Neuronal Culture Using a High-Density Neuronal Feeder Layer

2025

This video demonstrates the method for culturing ultra-low-density neurons in the presence of a high-density neuronal feeder layer. It establishes a co-culture of varying-density neurons, ensuring close physical proximity. The growth factors secreted by high-density neurons help neuronal survival and growth, maintaining ultra-low-density neurons for a longer time period.

This article describes the protocol for culturing low-density primary hippocampal neurons growing on glass coverslips inverted over a glial monolayer. The neuron and glial layers are separated by paraffin wax beads. The neurons grown by this method are suitable for high-resolution optical imaging and functional assays.

Low density cultures of primary hippocampal neurons usually require glia feeder layer to supply neurotrophic factors and sustain longevity. We describe here a simplified method to culture ultra-low density neurons on glass coverslips in the presence of a high density neuronal feeder layer, which facilitates investigation of specific neuronal-autonomous mechanisms.

Continuous Culture of Bacteria at Constant Density via Automated Growth Rate Estimation

Begin with a turbidostat, a device that maintains a constant cell density in culture. The device contains a vessel with a bacterial culture, which is continuously aerated by stirring and maintained at a constant temperature.A laser diode emits a light beam that is split into two paths. One path is directed to a reference sensor to monitor laser intensity fluctuations, while the other passes through the culture.Bacterial cells scatter and absorb light, decreasing the transmitted intensity, which...
