Human Endometrial Xenograft

A human endometrial xenograft is an experimental model in which human endometrial tissue is transplanted into an immunocompromised animal to study tissue behavior in a living organism. Because the host’s reduced immune response permits engraftment, transplanted endometrial cells can respond to hormonal signals, undergo cyclical changes, and interact with surrounding tissues. This model supports investigation of endometriosis, including lesion establishment, inflammation, vascularization, and hormone-dependent growth, while preserving tissue-level interactions that cell cultures may not reproduce. Human endometrial xenografts also help researchers evaluate disease mechanisms and assess potential therapeutic strategies before clinical studies.

This video demonstrates an assay to generate a humanized xenograft mouse model. An immunocompromised mouse is injected with cyclophosphamide to cause immune cell depletion. Patient-derived tumor cells mixed with human peripheral blood mononuclear cells (PBMCs) in a hydrogel are subcutaneously injected into the mouse, forming a tumor with infiltrating T cells.

Isolation of Stem Cells from Human Pancreatic Cancer Xenografts

Cancer stem cells (CSCs) have been identified in a number of malignancies. In this protocol we describe a flow cytometric method utilizing aldehyde dehydrogenase activity and CD44 and CD24 expression to isolate CSCs from human pancreatic adenocarcinoma xenografts. These viable cells can then be used in functional and analytical studies.

Human tumor xenografts in immunodeficient mice are valuable tools to study cancer biology. Specific protocols to generate subcutaneous and intrahepatic xenografts from human hepatocellular carcinoma cells or tumor fragments are described. Liver regeneration induced by partial hepatectomy in recipient mice is presented as a strategy to facilitate intrahepatic engraftment.

Mantle cell lymphoma (MCL) is a difficult to treat B cell disorder and it is equally difficult to establish a xenograft mouse model of primary MCL to study and develop therapeutics. Here, we describe the successful establishment of MCL xenografts in mice to help understand its underlying biology.
