Hybridization Ligation

Hybridization ligation is a molecular biology method that uses complementary nucleic acid pairing and enzymatic joining to detect, identify, or analyze specific DNA or RNA sequences. In the assay, oligonucleotide probes hybridize to adjacent regions of a target strand, positioning their ends for DNA ligase to seal the break when the sequence is correctly matched. This sequence-dependent ligation provides high specificity and can generate products for downstream detection, amplification, or analysis. The approach supports applications such as mutation and genotype discrimination, pathogen identification, gene analysis, and other studies requiring precise recognition of nucleic acid sequences.

Proximity Ligation Assay to Study In Situ Protein-Protein Interactions

This video describes the proximity ligation assay to study in situ protein-protein interactions in nematode cells. The close proximity of the proteins of interest generates the circular DNA template with the help of a PLA probe attached to a secondary antibody. The probe is further amplified using the amplification solution, which produces a long single-stranded DNA followed by the hybridization of the detector probe, enabling the localization of protein-protein interactions in cells.
