Hybridization Protocol

A hybridization protocol is a laboratory method for detecting or identifying a specific nucleic acid sequence by pairing it with a complementary DNA or RNA probe. The procedure involves preparing the target sample, allowing probe and target strands to anneal under controlled temperature, salt, and time conditions, and removing nonspecific interactions through stringency washes. A labeled probe then produces a measurable signal, often through fluorescence, chemiluminescence, or enzymatic detection. Hybridization protocols support Southern and Northern blotting, in situ hybridization, genotyping, gene-expression analysis, and localization of sequences within cells or tissues.

We describe a modified DIG in situ hybridization protocol, which is fast and applicable on a wide range of plant species including Norway spruce. With just a few adjustments, including altered RNase treatment and proteinase K concentration, the protocol may be used in studies of different tissues and species.
