Immunodetection

Immunodetection is a group of methods that uses antibodies to identify and localize specific proteins or other antigens in biological samples. The process relies on selective antigen-antibody binding, followed by detection of a label attached directly to the primary antibody or indirectly to a secondary antibody; fluorescent, enzymatic, or chemiluminescent signals reveal the target’s presence and distribution. Techniques such as immunoblotting, immunohistochemistry, and immunofluorescence apply this principle to analyze protein expression, cellular localization, and tissue organization. Immunodetection supports research in cell biology, pathology, microbiology, and biomedical science by providing qualitative and quantitative evidence about molecular changes.

Described here is a method to detect and quantify mitochondrial outer membrane proteins by immunolabeling of mitochondria isolated from rodent tissue and analysis by flow cytometry. This method can be extended to assess functional aspects of mitochondrial subpopulations.
