In Vitro Reconstitution

In vitro reconstitution is a method for rebuilding a biological process outside living cells by combining purified or defined components under controlled laboratory conditions. Researchers isolate the molecules required for an activity, such as proteins, nucleic acids, lipids, or energy sources, then adjust their concentrations, order of addition, temperature, and chemical environment to test how the system functions. This reductionist approach separates direct molecular interactions from the complexity of intact cells, helping identify mechanisms of processes such as membrane transport, cytoskeletal assembly, signaling, and gene expression. It also supports quantitative modeling, synthetic biology, and the design of controllable cell-free systems.

Efforts to isolate the catalytic subunit of telomerase, TERT, in sufficient quantities for structural studies, have been met with limited success for more than a decade. Here, we present methods for the isolation of the recombinant Tribolium castaneum TERT (TcTERT) and the reconstitution of the active T. castaneum telomerase ribonucleoprotein (RNP) complex in vitro.
