Hec1 Kinetochore Analysis

Hec1 kinetochore analysis is the study of Hec1, a core kinetochore protein, and its role in connecting chromosomes to spindle microtubules during cell division. As a component of the Ndc80 complex, Hec1 helps establish and regulate microtubule attachments at kinetochores, supporting chromosome alignment and accurate segregation. Researchers analyze Hec1 localization, abundance, interactions, or functional perturbation to assess kinetochore assembly and attachment stability. This approach is useful in cell biology and cancer research because defective Hec1-dependent connections can produce chromosome missegregation, aneuploidy, and compromised genome stability.

Here we report protocols to detect endogenous and exogenous centromere-kinetochore proteins in human cells and quantify these protein levels at centromeres-kinetochores by indirect immunofluorescent staining through the use of fixation (paraformaldehyde, acetone, or methanol fixation).

A method is described to visualize the recruitment and turnover of one of the kinetochore proteins and its coordination with the chromosome motion in Drosophila embryos using a Leica laser scanning confocal system.

This article describes a novel protocol and reagent set designed for sensitive measurement of neurotoxic effects of compounds and treatments on co-cultures of neurons and astrocytes using high content analysis. Results demonstrate that high content analysis represents an exciting novel technology for neurotoxicity assessment.
