Larval Body Wall Preparation

Larval body wall preparation is a dissection technique that exposes the muscles, nerves, and neuromuscular junctions of insect larvae for direct laboratory examination. Typically, an anesthetized larva is pinned in place, opened along the body wall, and cleared of internal tissues while the cuticle and musculature remain attached and gently stretched in physiological saline. This preparation preserves the organization of the larval nervous system and muscle fibers, allowing researchers to use microscopy, immunostaining, or electrophysiology to study synaptic structure and function. In biology, it supports investigations of neural development, motor control, synaptic plasticity, and genetic effects on neuromuscular organization.

Drosophila melanogaster larvae provide an ideal model system to investigate the mechanisms of axonal transport within larval segmental nerves. Using this procedure, 3rd instar larvae carrying various mutations can be compared to wild type larvae.

To understand how complex cell shapes, such as neuronal dendrites, are achieved during development, it is important to be able to accurately assay microtubule organization. Here we describe a robust immunohistological labeling method to examine microtubule organization of dendritic arborization neuron sensory dendrites, trachea, muscle, and other Drosophila larva body wall tissues.
