Lentiviral Reporter

A lentiviral reporter is a gene-delivery system that uses a lentiviral vector to introduce a detectable reporter gene, such as GFP or luciferase, into target cells, enabling researchers to monitor cellular events. After entry, the vector undergoes reverse transcription, and its genetic cargo can integrate into the host-cell genome, allowing sustained reporter expression in both dividing and nondividing cells. In biology, lentiviral reporters support visualization and quantification of gene expression, promoter activity, cell lineage, infection, and experimental perturbations. Their stable signal makes them useful for tracking cellular behavior over time and evaluating molecular mechanisms in research models.

A Lentiviral Transcriptional Reporter System to Generate a Glioblastoma Stem Cell Differentiation Reporter Line

2025

This video demonstrates a technique to generate a glioblastoma stem cell reporter line to identify the differentiation of glioblastoma stem cells (GSCs) into astroglia. It outlines the steps involved in seeding GSCs, transducing them with a lentivirus encoding GFP, and determining GFP reporter expression through flow cytometry.
