Lithium Acetate Method

The lithium acetate method is a chemical transformation technique used to introduce foreign DNA into yeast cells, enabling genetic manipulation in biological research. In this process, lithium ions and polyethylene glycol help make the yeast cell wall and membrane more permissive to DNA uptake, while heat shock can promote entry of the DNA into the cell. Transformed cells are then cultured on selective media so only those carrying the introduced genetic material grow. The method supports gene function studies, protein production, strain engineering, and analysis of cellular pathways, making it a widely used and accessible approach in molecular biology.

Direct Detection of the Acetate-forming Activity of the Enzyme Acetate Kinase

A method for the determination of acetate kinase activity is described. This assay utilizes a direct reaction for determining enzyme activity and kinetics of acetate kinase in the acetate-forming direction with different phosphoryl acceptors. Furthermore, this method can be utilized for assaying other acetyl phosphate or acetyl-CoA utilizing enzymes.

We describe the design and construction of an electrochemical cell for the examination of electrode materials using in situ neutron powder diffraction (NPD). We briefly comment on alternate in situ NPD cell designs and discuss methods for the analysis of the corresponding in situ NPD data produced using this cell.
