Lung Transplantation

Lung transplantation is a surgical procedure that replaces one or both diseased lungs with donor lungs to restore respiratory function when severe lung disease no longer responds to conventional treatment. During transplantation, surgeons connect the donor airways and blood vessels to the recipient’s remaining structures, while immunosuppressive medicines reduce T-cell-mediated recognition and rejection of the transplanted tissue. The procedure can improve oxygen exchange, exercise capacity, and quality of life for carefully selected patients with advanced pulmonary disorders. However, long-term outcomes depend on donor and recipient compatibility, infection prevention, monitoring for acute or chronic rejection, and management of complications, making transplantation an important intersection of surgery, immunology, and respiratory biology.

Here, we describe the necessary steps for establishing a rat EVLP model and show the inflammatory profile associated with the perfused lungs. The aim is to propagate knowledge and experiences about the rat EVLP model, enabling the integral understanding of the biological responses associated with that revolutionary technique.

Obliterative bronchiolitis is the key impediment to the long-term survival of lung transplant recipients and the lack of a robust preclinical model precludes examining obliterative bronchiolitis immunopathogenesis. Unlike other solid organ transplants, vascularized mouse lung transplantation has only recently been developed. Here we show our independently developed obliterative bronchiolitis model after murine orthotopic single-lung transplantation.

Rat Heterotopic Abdominal Heart/Single-lung Transplantation in a Volume-loaded Configuration

We describe a novel technique for heterotopic abdominal heart-lung transplantation (HAHLT) in rats. The transplant configuration results in a partially loaded graft circulation, allowing direct functional assessment. This model may be employed for acute or chronic studies of function and immunologic status of the transplanted graft.
