Wolbachia Tissue Crossing

Wolbachia tissue crossing is the movement of intracellular Wolbachia bacteria between host tissues, a process that shapes where this symbiont persists and how it is transmitted. Because Wolbachia resides within host cells, crossing tissue boundaries depends on infected-cell movement or division, bacterial release and uptake, and host membrane and cytoskeletal processes that support intracellular trafficking. Understanding these routes helps explain Wolbachia distribution in insects, including access to reproductive tissues and other organs, and clarifies effects such as maternal transmission and reproductive manipulation. This knowledge supports studies of host–microbe biology and Wolbachia-based strategies for managing insect-borne disease.

Aedes aegypti mosquitoes infected with Wolbachia are being released into natural populations to suppress the transmission of arboviruses. We describe methods to rear Ae. aegypti with Wolbachia infections in the laboratory for experiments and field release, taking precautions to minimize laboratory adaptation and selection.
