Media Flow Rate

Media flow rate is the volume of culture medium moving through a biological system per unit of time, an important parameter for maintaining suitable conditions for cells and tissues. In perfusion cultures and bioreactors, pumps regulate this rate to control the delivery of nutrients and oxygen, removal of metabolic waste, and fluid-induced shear stress experienced by cells. Adjusting media flow rate helps researchers balance cell growth, viability, and tissue development while preventing nutrient depletion or excessive mechanical stimulation. This measurement supports the optimization of cell culture, tissue engineering, and other biological systems that require controlled fluid movement.

2023

Source: Ricardo Mejia-Alvarez and Hussam Hikmat Jabbar, Department of Mechanical Engineering, Michigan State University, East Lansing, MI The purpose of this experiment is to demonstrate the calibration of a flow passage as a flowmeter using a control volume (CV) formulation [1, 2]. The CV analysis focuses on the macroscopic effect of flow on engineering systems, rather than the detailed description that could be achieved with a detailed differential analysis. These two techniques should be...

2025

Source: Jagau, H. et al. Pneumococcus Infection of Primary Human Endothelial Cells in Constant Flow. J. Vis. Exp. (2019)This video demonstrates the real-time visualization of pneumococcal attachment to Von Willebrand Factor (VWF) strings on endothelial cells under constant media flow conditions. It outlines the steps for stimulating VWF release, labeling the strings, and quantifying bacterial binding.

The protocol describes a reproducible method designed for use with cell culture supernatants to detect surface epitopes on small extracellular vesicles (EV). It utilizes specific EV immunoprecipitation using beads coupled with antibodies that recognize surface antigen CD9, CD63, and CD81. The method is optimized for downstream flow cytometry analysis.

This video describes the rheotaxis assay in zebrafish. The method involves the measurement of orientation behavior of zebrafish in response to different water flow rates under the influence of different magnetic fields.

Renal Drug Excretion: Effect of Urine pH, Flow Rate, and Drug pKa

2025

The pH of urine, the drug's pKa, and the urine flow rate are vital parameters for drug reabsorption and excretion. Urinary pH varies between 4.6 and 8.0 and is influenced by diet, drug intake, and the patient's pathophysiology. It affects a drug's ionization state and reabsorption. For instance, carbohydrate-rich food produces alkaline urine promoting drug excretion, while proteins and certain medications like ascorbic acid lead to acidic urine enhancing reabsorption. The pKa of a drug,...
