Skin Derived Precursor Cells

Skin-derived precursor cells (SKPs) are multipotent cells isolated from adult skin, particularly the dermis, with the capacity for self-renewal and differentiation into several cell types. Researchers typically enrich SKPs by culturing dissociated skin cells in serum-free, growth factor–supplemented conditions, where they form free-floating spheres and can later differentiate into neural, glial, smooth muscle, or adipocyte-like cells. In biology, SKPs provide a model for studying cell fate, tissue development, and regenerative potential. Their accessibility from skin also supports research into disease modeling, nervous system repair, and strategies for generating specialized cells for tissue engineering.

Differentiation of Embryonic Stem Cells into Oligodendrocyte Precursors

We describe a small molecule-based protocol for differentiation of mouse embryonic stem cells into oligodendrocyte precursor cells (OPCs). This protocol generates Olig2+NG2+ OPCs with high efficiency by 30 days of differentiation. We also describe a method to generate "spiking" OPCs that can fire action potentials.
