Melting Curve Analysis

Melting curve analysis is a technique used to characterize double-stranded DNA by monitoring how it separates as temperature increases. In real-time PCR, a fluorescent dye or probe reports the loss of double-stranded DNA, producing a fluorescence-versus-temperature profile whose peak identifies the melting temperature (Tm), the point at which the DNA molecules denature. Because Tm depends on sequence composition, length, and complementary pairing, even a mismatch can alter the curve. Researchers use melting curve analysis to assess PCR product specificity, distinguish closely related sequences, and support genotyping or mutation screening without sequencing, making it a rapid quality-control and analytical tool in molecular biology.
