Metal-limited Growth Assay

A Metal-limited Growth Assay is a biological method that measures how restricted availability of an essential metal affects the growth of cells or microorganisms. In this assay, cultures are exposed to controlled metal concentrations, often across a gradient, and growth is tracked by changes in cell density or another quantitative signal. Comparing growth under metal-replete and metal-limited conditions reveals metal requirements, limitation thresholds, and responses to deficiency. The approach supports studies of nutrient physiology, metal homeostasis, microbial ecology, and host-microbe interactions, while also helping evaluate how environmental metal availability influences biological activity.

We describe here a method for growth of Neisseria gonorrhoeae in metal-restricted liquid medium to facilitate the expression of genes for metal uptake. We also outline downstream experiments to characterize the phenotype of gonococci grown in these conditions. These methods may be adapted to be suitable for characterization of metal-responsive genes in other bacteria.
