Microsatellite Amplification

Microsatellite amplification is a molecular biology method used to copy short, tandemly repeated DNA sequences so their length variation can be measured among individuals or samples. The process typically uses polymerase chain reaction (PCR), in which primers flank the repeat region and cycles of denaturation, primer annealing, and DNA synthesis generate millions of target copies; fragment sizes are then separated by electrophoresis or analyzed with fluorescent detection. Because microsatellites are highly variable, amplification supports genetic profiling, population studies, linkage mapping, biodiversity assessments, and forensic identification. Reliable results depend on specific primers and careful control of nonspecific products and allele sizing.
