Mitochondrial Isolation

Mitochondrial isolation is a laboratory method for separating mitochondria from cells or tissues so their structure, function, and biochemical activity can be studied independently. The process typically uses controlled homogenization to break open cells while preserving organelles, followed by differential centrifugation, in which sequential centrifugal forces separate mitochondria from nuclei, membranes, cytosol, and other components according to size and density. Isolated mitochondria support research on cellular respiration, ATP production, membrane potential, metabolism, and apoptosis. The quality of the preparation depends on maintaining appropriate temperature, osmotic conditions, and gentle handling, making the technique valuable in physiology, disease research, toxicology, and drug development.

Mitochondrial Isolation from Skeletal Muscle

This protocol describes a procedure to study the respiration of mitochondria isolated from skeletal muscles. This method was adapted from Scorrano et al. (2007). The mitochondrial isolation procedure requires about 2 hours. The mitochondrial respiration can be completed in about 1 hour.
