Induced Pluripotent Stem Cells

Induced pluripotent stem cells (iPSCs) are mature, specialized cells that have been genetically reprogrammed to return to a pluripotent state, enabling them to produce many cell types in the body. Researchers typically introduce reprogramming factors that alter gene expression and reset the cell’s developmental identity, often converting adult somatic cells such as skin or blood cells into stem-like cells. In biology, iPSCs provide patient-specific models for studying development and disease, testing drug responses, and investigating regenerative medicine. Because they can be generated without using embryonic tissue, they also support personalized research while raising important questions about genetic stability and safety.

Induced Pluripotent Stem Cells

Stem cells are undifferentiated cells that divide and produce different types of cells. Ordinarily, cells that have differentiated into a specific cell type are post-mitotic—that is, they no longer divide. However, scientists have found a way to reprogram these mature cells so that they “de-differentiate” and return to an unspecialized, proliferative state. These cells are also pluripotent like embryonic stem cells—able to produce all cell types—and are therefore called induced pluripotent stem...

Despite recent advancements in genetic modification, transfection of human embryonic stem cells (HESCs) remains a capricious process. To our knowledge, systematic and efficient methods to transfect human induced pluripotent stem cells (iPSCs) have not been reported. Here, we describe robust protocols to efficiently transfect and nucleofect human iPSCs.

Generating induced pluripotent stem cell (iPSC) lines produces lines of differing developmental potential even when they pass standard tests for pluripotency. Here we describe a protocol to produce mice derived entirely from iPSCs, which defines the iPSC lines as possessing full pluripotency1.

A method to generate human induced pluripotent stem cells (iPSCs) via retrovirus-mediated ectopic expression of OCT4, SOX2, KLF4 and MYC is described. A practical way to identify human iPSC colonies based on GFP expression is also discussed.

Differentiation of Induced Pluripotent Stem Cells into Neural Progenitor Cells

The video demonstrates a technique for differentiating human induced pluripotent stem cells (hiPSCs) into neural progenitor cells (NPCs). Initially, the hiPSCs are cultured on a feeder layer of mouse embryonic fibroblasts (MEFs). Subsequently, the cells are detached and transferred to a biopolymer-coated plate to allow the MEFs to adhere to and separate from the hiPSCs. The suspended hiPSCs are then transferred to a V-bottom plate and centrifuged to form aggregates. Finally, the addition of a...
