Morphology Assay

A morphology assay is a biological method that evaluates the form, structure, and spatial organization of cells, tissues, or organisms to detect phenotypic changes. It typically combines microscopy with image analysis, using staining or fluorescent labeling to visualize samples and measurements such as cell area, shape, nuclear features, or tissue organization to quantify differences. In biology, morphology assays help assess cell growth, differentiation, migration, viability, and responses to drugs or environmental conditions. By converting structural observations into measurable data, they support disease modeling, toxicity testing, developmental studies, and investigations of cellular function.

Rectal Organoid Morphology Analysis (ROMA): A Diagnostic Assay in Cystic Fibrosis

2022

This protocol describes rectal organoid morphology analysis (ROMA), a novel diagnostic assay for cystic fibrosis (CF). Morphological characteristics, namely the roundness (circularity index, CI) and the presence of a lumen (intensity ratio, IR), are a measure of CFTR function. Analysis of 189 subjects showed perfect discrimination between CF and non-CF.

Bacterial and archaeal cells exhibit remarkable diversity in shape and structure, critical in their adaptability and functionality. Among bacteria, the most commonly observed shapes include cocci and bacilli. Cocci are spherical and may exist singly or in groupings such as pairs (diplococci), chains (streptococci), clusters (staphylococci), or tetrads. Bacilli, in contrast, are rod-shaped and can also occur as single cells, in pairs, or chains, depending on their environmental and genetic...

Two-Photon Microscopy for Imaging Neuronal Morphology in a Mouse Pup

Source: Mizuno, H., et al. In Vivo Two-photon Imaging of Cortical Neurons in Neonatal Mice. J. Vis. Exp. (2018)This video showcases two-photon microscopy being used to image neurons in the brain of an anesthetized transgenic mouse pup. The two-photon laser allows for deeper imaging with high spatial resolution and minimal light scattering, enabling the detailed capture of neuronal morphology, including dendritic projections.
