Trypanosome Editosome

The trypanosome editosome is a multiprotein complex that modifies mitochondrial RNA transcripts in kinetoplastid parasites, enabling the expression of genes that would otherwise remain incomplete or unreadable. Guided by complementary guide RNAs, the editosome cleaves precursor transcripts, inserts or removes uridine residues, and then processes and ligates the edited RNA into a functional message. This specialized form of RNA editing is essential for mitochondrial gene expression and parasite survival. Studying editosome components and their coordinated enzymatic activities provides insight into RNA processing, mitochondrial biology, and parasite-specific molecular mechanisms, while identifying potential targets for therapies against trypanosomiasis.

A highly sensitive ribozyme-based assay, applicable to high-throughput screening of chemicals targeting the unique process of RNA editing in trypanosomatid pathogens, is described in this paper. Inhibitors can be used as tools for hypothesis-driven analysis of the RNA editing process and ultimately as therapeutics.

Glycosome dynamics in African trypanosomes are difficult to study by traditional cell biology techniques such as electron and fluorescence microscopy. As a means of observing dynamic organelle behavior, a fluorescent-organelle reporter system has been used in conjunction with flow cytometry to monitor real-time glycosome dynamics in live parasites.

This method of trypanosome separation from blood depends on their surface charge being less negative than mammalian blood cells. Infected blood is placed and treated on an anion-exchanger column. This method, the most fitting diagnostic for African trypanosomiasis, provides purified parasites for immunological, biological, biochemical, pharmaceutical and molecular biology investigations.
