Mutant Line Screening

Mutant line screening is the systematic examination of genetically altered organisms or cell lines to identify heritable changes in traits, gene function, or biological responses. Researchers compare mutant lines with appropriate wild-type controls under controlled conditions, using phenotypic measurements and, when needed, genotyping to link observed differences to specific genetic changes. In biology, this approach helps reveal how genes influence development, physiology, behavior, and disease-related processes. Screening can also identify lines with useful characteristics for functional genomics, model-organism research, and breeding, providing a foundation for investigating gene mechanisms and selecting candidates for further study.

We describe a streamlined protocol for generating "fillet" preparations of Drosophila embryos of specific genotypes. This protocol allows efficient execution of a variety of genetic screens. It also allows excellent visualization of structures in the late embryo.
