Mutation Detection

Mutation detection is the identification of changes in an organism’s DNA sequence, an essential process for understanding genetic variation, disease, and biological inheritance. It works by comparing DNA from a sample with a reference sequence or control, using approaches such as polymerase chain reaction, DNA sequencing, hybridization assays, or targeted variant analysis to locate substitutions, insertions, and deletions. In biology, mutation detection supports studies of gene function, population diversity, cancer-associated variants, inherited disorders, and pathogen evolution. Accurate detection also helps researchers connect specific sequence changes with molecular and cellular effects.

Wild-Type Blocking PCR to Detect Low-Frequency Somatic Mutations

This video describes wild-type blocking polymerase chain reaction, or WTB-PCR, which detects somatic mutations. The PCR-based technique uses a locked nucleic acid, or LNA, an oligonucleotide that binds to its complementary wild-type allele and blocks its elongation. The blocking selectively allows the amplification of low-level mutant alleles over their wild-type variants in the sample.
