Nadh Oxidation

NADH oxidation is the biological process in which reduced nicotinamide adenine dinucleotide (NADH) donates electrons and is converted back to NAD+, a reaction essential for maintaining cellular energy metabolism. In aerobic respiration, NADH transfers electrons to the mitochondrial electron transport chain, where their stepwise movement through protein complexes drives proton pumping across the inner mitochondrial membrane and helps establish the gradient used to produce ATP. NADH oxidation also supports the continued operation of glycolysis, the citric acid cycle, and other pathways that rely on NAD+ as an electron acceptor. Measuring this process helps researchers study cellular respiration, metabolic regulation, and mitochondrial function.

The objective is to monitor the mitochondrial redox state of isolated hearts within the context of physiologic preload and afterload pressures. A biventricular working rabbit heart model is presented. High spatiotemporal resolution fluorescence imaging of NADH is used to monitor the mitochondrial redox state of epicardial tissue.

Here we describe a method to directly visualize microregional tissue hypoxia in the mouse cortex in vivo. It is based on concurrent two-photon imaging of nicotinamide adenine dinucleotide (NADH) and the cortical microcirculation. This method is useful for high resolution analysis of tissue oxygen supply.
