Necrosis Assessment

Necrosis assessment is the evaluation of cell death marked by loss of plasma-membrane integrity and release of intracellular contents, providing evidence of tissue injury or cellular damage. It typically combines morphological examination with assays that detect membrane disruption, cytoplasmic leakage, or changes in cellular viability, distinguishing necrotic cells from healthy or apoptotic cells when appropriate. In biology, these assessments help characterize responses to toxins, infection, ischemia, physical stress, and experimental treatments. Reliable measurement supports studies of tissue pathology, inflammation, disease mechanisms, and therapeutic safety, while enabling researchers to compare how biological conditions alter cell survival and damage.

Necrosis is considered as an “accidental” or unexpected form of cell death that ends in cell lysis. The first noticeable mention of “necrosis” was in 1859 when Rudolf Virchow used this term to describe advanced tissue breakdown in his compilation titled “Cell Pathology”. Morphological Manifestations of Necrosis Necrotic cells show different types of morphological appearance depending on the type of tissue and infection. In coagulative necrosis, cells become anucleated and die, but their...

Muscle Necrosis Assay in C. elegans Induced by a Pathogenic Aeromonas Strain

Source: Chen, Y. et. al., Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018)This video demonstrates a muscle necrosis assay in C. elegans (Caenorhabditis elegans) using pathogenic Aeromonas strains. By feeding green fluorescent protein (GFP)-expressing worms on Aeromonas-seeded plates, the assay models bacterial infection and reveals tissue damage through fluorescence imaging. The progressive loss of muscle structure and GFP...

Cellular Injury IV: Necrosis

Necrosis is a form of irreversible cell death caused by severe injury such as ischemia, toxins, or trauma. Unlike programmed cell death, it is an uncontrolled, pathological process that typically provokes inflammation in surrounding tissues.Pathophysiologic ChangesNecrosis begins when cells sustain critical damage, leading to swelling of organelles, particularly mitochondria, and rapid ATP depletion. As energy levels decline, membrane ion pumps fail, leading to calcium influx and eventually,...

The primary outcome measure in clinical trials for neuromuscular disorders is generally improved muscle function. Therefore, assessing the effect of potential therapeutic compounds on muscle performance pre clinically in mouse models is of great importance. We here describe several functional tests to address this.
