Neural Fold Grafting

Neural fold grafting is an embryological transplantation technique used to examine how tissues at the edges of the neural plate contribute to nervous system development. During the procedure, a neural fold or portion of it is microsurgically removed from one embryo and placed into a corresponding site in another, often using tissue labels to track cell movements and fate. As development proceeds, grafted cells may integrate, migrate, or differentiate according to their origin and new environment. This method provides direct evidence for neural crest behavior, neural tube formation, and the development of craniofacial structures and the peripheral nervous system.

This video illustrates a technique for isolating cranial neural folds from the midbrain region of chick embryos. It presents a detailed dissection process of the neural folds, their placement, and attachment on fibronectin-coated coverslips, followed by the observation of neural crest cell migration from the neural folds.

Here we report dual labeling of neural crest cells and blood vessels using chickGFP neural tube intraspecies grafting combined with intra-vascular DiI injection. This experimental technique allows us to simultaneously visualize and study development of the NCC-derived (enteric) nervous system and the vascular system, during organogenesis.
