Oil Mounting Technique

The oil mounting technique is a microscopy method that places a biological specimen in a clear, nonaqueous oil-based medium between a slide and coverslip for detailed observation. The mounting medium surrounds and stabilizes the specimen while reducing light scattering and improving optical clarity, particularly when its refractive index more closely matches that of glass and the sample. This approach can preserve structural features for examination under bright-field or other light microscopy methods, supporting the study of cells, tissues, microorganisms, and prepared biological specimens. Proper mounting also helps limit movement, maintain specimen integrity, and produce clearer, more consistent microscopic images.

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.
