Oligosaccharide Analysis

Oligosaccharide analysis is the characterization of short carbohydrate chains, including their composition, sequence, branching, and linkage patterns, which influence molecular recognition and biological function. Researchers typically release or isolate oligosaccharides from glycoconjugates, separate them by liquid chromatography, and determine their masses and structures using mass spectrometry, often with enzymatic digestion or chemical derivatization to distinguish isomers and glycosidic linkages. In biology, these analyses clarify cell-surface glycans, protein glycosylation, host–pathogen interactions, and changes associated with disease or development. The resulting structural data support biomarker discovery, glycoengineering, and the development of carbohydrate-based therapeutics and diagnostics.

We describe a method for analysis of the alteration of N-linked glycans through the early life of glycoproteins after their biosynthesis in mammalian cells. This is achieved by pulse-chase analysis of metabolically labeled glycans, enzymatic release from glycoproteins and examination by HPLC.

Mass Spectrometric Analysis of Glycosphingolipid Antigens

A specific and sensitive method to gain insight into the expression profile of glycosphingolipid antigens in immune organs and cells is described. The method takes advantage of the ion trap mass spectrometry allowing step-wise fragmentation of glycosphingolipid molecules for structural analysis in comparison to chemically synthesized standards.

Analysis of SCAP N-glycosylation and Trafficking in Human Cells

We describe a modified method for membrane fraction isolation from human cells and sample preparation for the detection of SCAP N-glycosylation and total protein by using western blot. We further introduce a GFP-labeling method to monitor SCAP trafficking using confocal microscopy. This protocol can be used in regular biology laboratories.
