Optogenetic Stimulation

Optogenetic stimulation is a biological technique that uses light-sensitive proteins to control the activity of specific cells with high temporal and spatial precision. Researchers introduce microbial opsins, such as light-gated ion channels or pumps, into target cells, then deliver selected wavelengths of light to alter membrane potential and trigger or inhibit cellular signaling. In biology, this approach enables controlled studies of neural circuits, muscle activity, sensory processing, and behavior in living organisms. By linking light delivery to cell-type-specific genetic targeting, optogenetics helps reveal causal relationships between cellular activity and physiological function, while supporting research into disease mechanisms and potential therapeutic strategies.

Genetically encoded optogenetic tools enable noninvasive manipulation of specific neurons in the Drosophila brain. Such tools can identify neurons whose activation is sufficient to elicit or suppress particular behaviors. Here we present a method for activating Channelrhodopsin2 that is expressed in targeted neurons in freely walking flies.

This video demonstrates a study of the navigational behavior of Drosophila larvae in response to optogenetic stimulation of olfactory sensory neurons (ORNs). Transgenic Drosophila larvae expressing light-sensitive ion channels in the ORNs are placed on agarose within a behavior arena. Optogenetic stimulation with light opens the channels, causing a cation influx that triggers the transmission of the signal to the brain, and the navigation behavior is recorded.

Cochlear implants (CIs) enable hearing by direct electrical stimulation of the auditory nerve. However, poor frequency and intensity resolution limits the quality of hearing with CIs. Here we describe optogenetic stimulation of the auditory nerve in mice as an alternative strategy for auditory research and developing future CIs.
