Organoid-derived Model

An organoid-derived model is an experimental system generated from three-dimensional organoids to reproduce selected structural, cellular, or functional features of a tissue in the laboratory. Organoids typically form when stem or progenitor cells are maintained under defined culture conditions that support cell proliferation, differentiation, and self-organization; researchers can then use the resulting tissue-like structures or their derived cells for controlled experiments. In biology, these models help investigate development, tissue function, disease mechanisms, and responses to genetic or pharmacological perturbations. They provide a bridge between simplified cell cultures and complex organisms, while supporting reproducible studies of human-relevant biology.

Here, we describe the preparation of human organoid-derived intestinal epithelial monolayers for studying intestinal barrier function, permeability, and transport. As organoids represent original epithelial tissue response to external stimuli, these models combine the advantages of expandability of cell lines and the relevance and complexity of primary tissue.

This protocol allows for the differentiation of human pluripotent cells into intestinal organoids. The protocol mimics normal human development by differentiating cells into a population of definitive endoderm, hindgut endoderm and then intestinal epithelium. This makes the protocol suitable for studying both intestinal development as well as disease modelling applications.

Patient-derived organoid cultures of pancreatic ductal adenocarcinoma are a rapidly established 3-dimensional model that represent epithelial tumor cell compartments with high fidelity, enabling translational research into this lethal malignancy. Here, we provide detailed methods to establish and propagate organoids as well as to perform relevant biological assays using these models.

Here, we present a protocol to generate melanoma patient-derived organoids by culturing disassociated cell suspensions from fresh melanoma tissues. These organoids faithfully recapitulate patient-specific tumors in vitro, offering an innovative approach to exploring tumor immunosuppressive mechanisms, drug screening, drug resistance mechanisms, and cancer surveillance approaches.
